I know how to do it, all I'm asking is do the files need to be the EXACT same size as what I am replacing?

No they don't. But inserting larger tunes will mean you will need to enlarge your O_SOUNDS.AFS and then regenerate your PES5 ISO.

DkZ Studio is great for recreating AFS files and there are guides available for creating new ISO's containing larger files. Good luck!
