If you do have the privilege of playing online, you will soon notice that 70% of online users will go for Barcelona or Madrid even if their username is "Milanforlife". It is quite frustrating indeed, but whatever you do please DO NOT pick madrid\barsa just to match them. It is pointless and not as rewarding as beating those users with say, Arsenal or Everton.
After 100+ games online and probably 80 games were against barsa\madrid, here are some tips on how to maintain a clean sheet against them:
1) Never ever play with a high line, this aint PES2011 and if Messi breaks through, most likely you will be chasing his shadow hoping he wont score. I lost 5-0 with a high line and 0-0 with a deep line. Obviously its crucial
2) Set the defense line to "deep", I use around 3\20. This way your box will be full of players.
3) Set the compactness to "narrow", I use 5\20. This means your players will stay together and very compact. Useful if ur opponents just runs with ronaldo\messi down the wings looking for a cross and tap in. Crossing is very easy in this game so having extra men in the box is good insurance.
4) While facing speedy players, do not challenge him right away. use the jockey button and hold ur ground. Jockey + tackle is useful if you know how to use it correctly and most of the time ur defender will tackle on its own when opponents gets too close. It also automatically blocks shots. Do not over use it though.
5) Have strong midfielders chasing people down but maintaining ur back 4 ready for lob balls. I play with Arsenal, therefore I usually have song as DM, DIABY as CM and Wilshire as an creative CM.
6) Frustrate them with Arsenal style football, pass pass pass pass and pass. Soon they will be so angry that often leads to mistakes.
Most of my games have ended 1-0 or 0-0 or 1-1. The 1.02 has helped a lot since tackling is better and gameplay is a bit slower. The only times I get completely destroyed are when i have been playing way too long and losing focus. FOCUS IS CRUCIAL when defending against these barsa\madrid users. A simple ball not picked up can mean 1-0 down.
