Are there enough differences and updates in PES6 and 5 to warrant upgrading. I have PES4 at the moment, which i enjoy greatly. Is 6 and or 5 that superior to it?

Aside from the transfers update, it's very hard to tell if the gameplay is better.
I think it will be fair to say that the graphics are better, but after playing a lot of PES4, PES5 and PES6 I can say that:
  • I could hardly notice any difference in difficulty, AI or gameplay between 5 and 6.
  • I did noticed that 5 is harder than 4, although it has some annoying stuff that weren't around in 4. Although it's been a long time since I played 4 so I don't really rememebr what it's like now.
  • All in all, when I think about it I guess that the gameplay is somewhat better and more realistic in 5 than in 4.

All of this is reagrding the PC versions.
